简要介绍:

科研工作主要集中于动物基因修饰和基因敲除、人和小鼠胚胎干细胞基因操作和定向分化、家畜动物胚胎干细胞的分离和培养等方面。用经典的同源重组方法将绿色荧光蛋白(GFP)和红色荧光蛋白(RFP)基因定点整合到小鼠胚胎干细胞的胰-十二指肠同源盒1(pancreatic and duodenal homeobox 1,PDX1)基因和胰岛素(InsulinINS)基因位点,在体外定向分化过程中通过检测绿色荧光蛋白和红色荧光蛋白的表达以确定胚胎干细胞向胰岛β-细胞分化,用以鉴定胚胎干细胞定向分化的生长因子以及寻找胚胎干细胞向β-细胞定向分化的最佳方法,此研究结果发表于《Stem Cell Research》。首次在人的胚胎干细胞的胰岛素基因位点定点插入绿色荧光蛋白基因,而且在体外分化的β-细胞中都检测到绿色荧光蛋白的表达,证明绿色荧光蛋白忠实地代表了胰岛素基因的表达,可以用于代替胰岛素基因来分离和鉴定β-细胞,解决了免疫荧光检测的繁琐步骤,而且可以用流式细胞分离仪轻松分离β-细胞,进行进一步的生长成熟的研究,大大促进了胚胎干细胞对于糖尿病的治疗研究,此研究结果发表在《Diabetologia》。目前主要从事最新基因编辑技术应用于草食家畜的基因编辑,以及牛羊胚胎干细胞的建立方面的研究。

    本人或与他人合作在国际和国内科研杂志上发表学术论文20多篇,其中10多篇为SCI収录。
